日前,位于合肥高新區(qū)的安徽中盛溯源生物科技有限公司研發(fā)的“NCR100注射液”獲批臨床,擬開展針對膝骨關(guān)節(jié)炎的臨床試驗(yàn),這也是國內(nèi)首款獲批臨床的誘導(dǎo)多能干細(xì)胞(iPSC)衍生的間充質(zhì)干細(xì)胞(iMSC)藥物[1]。

膝關(guān)節(jié)炎是中老年人常見的關(guān)節(jié)退行性病變。骨關(guān)節(jié)炎是以關(guān)節(jié)軟骨破壞及關(guān)節(jié)邊緣和軟骨下骨質(zhì)再生為特征的慢性關(guān)節(jié)疾病。本病病理核心是關(guān)節(jié)軟骨的損傷及骨贅形成。目前認(rèn)為本病是多因素聯(lián)合作用的結(jié)果:軟骨基質(zhì)合成與分解代謝失調(diào),軟骨下骨板損害使軟骨失去緩沖作用,關(guān)節(jié)內(nèi)局灶性炎癥[2]。臨床常見患者膝關(guān)節(jié)的疼痛、晨僵、腫脹、摩擦音及畸形,嚴(yán)重影響了中老年人的生活質(zhì)量。

一些因子抑制半胱氨天冬氨酸蛋白酶的裂解從而參與炎癥調(diào)節(jié)過程。iPSC-MSCs已經(jīng)成為骨髓源、臍帶源等MSCs和再生藥物治療骨關(guān)節(jié)炎的一種很有前途的替代細(xì)胞來源[8]。iPSC-MSCs治療骨關(guān)節(jié)炎的研究很多,其療效,副作用及其作用機(jī)制也在進(jìn)一步確認(rèn)和深入研究中。
